T-Cell / T-Lymphopoietic Progenitor Cells


T-cells are not supposed to play an important role unless unless the body recognizes an infection or other perturbation that will induce the production of lymphopoietic stem cells and, in turn, the production of T-cell or T-lymphocyte progenitor cells.

Although T-cell progenitors can be stimulated to a low degree with species-specific interleukin-2 (IL-2), co-factors are usually needed to allow the necessary T-cell interactions to take place. As such, Preferred Cell Systems has colony-forming cell reagents and assays that include IL-2, but also need the addition of specific co-stimulators.

More recently, Preferred Cell Systems™ has developed T-cell expansion reagents and highly sensitive and advanced, instrument-based assays to quantitatively measure T-cell progenitor stimulation for various applications. These reagents and assays can now be obtained with species-specific co-stimulators included in the Suspension Expansion Culture™ (SEC™) Master Mix.

The assays, in particular, have been designed so that not only the proliferation status of the cells can be measured, by the type of T-cells produced can be identified using phenotypic analysis by flow cytometry. Indeed, these assays can be combined or multiplexed with several different additional readouts.

Preferred Cell Systems™ distinguishes between T-cell progenitors cultured using SEC™ Technology and those grown in methylcellulose. All T-cell progenitor using SEC™ technology are designed with the prefix "P" for "Progenitor".

In the table below, you will find the T-cell progenitor populations developed by Preferred Cell Systems™ that can be detected using either a SEC™-based or a methylcellulose assay. These are:
  • CellExpand = Flexible readout of cell expansion
  • HALO = Bioluminescence readout
  • HemoFLUOR = Fluorescence readout
  • HemoLIGHT = Absorbance/colorimetric readout
  • ColonyGro & CAMEO-4 = Manual or automated colony counting
  • CAMEO-96 = Manual or automated colony counting following by bioluminescence measurement
The type of assay you choose is dependent upon a number of factors, that include, but are not limited to:
  • Use or application goal (research, cellular therapy or toxicity testing).
  • Whether you need to measure proliferation, differentiation or both. 
  • Cell type being detected.
  • Instrumentation available.
  • Other factors such as accuracy, sensitivity, precision etc.
